effects of gene polymorphisms in tumor necrosis factor-α on the susceptibility of type I diabetes mellitus in an Iraqi population

Background: Type 1 diabetes mellitus (T1DM) is a chronic, immune-mediated disease characterized by the destruction of insulin-producing β cells in pancreas. This study investigated influence single nucleotide polymorphisms (SNP) tumor necrosis factor (TNF-α) –308 G/A (rs1800629,) on T1DM patients. Method: The was included 75 patients and 25 healthy subjects. SNP–308 TNF-α gene detected using polymerase chain reaction restriction fragment length polymorphism (RFLP-PCR) technique controls. Results: genotype distribution results -308G/A SNP Tumor Necrosis Factor - α (TNF- α)-308 showed significant difference (p<0.05) between controls (GG: n= 24, 96%; GA: n=1, 4%; AA: n=0, 0%) 44, 58.67%; n=21, 28%; n=10, 13.33%).These an increased GA, AA G allele TNF- α(-308) than controls, they were significantly more likely to have mutant (OR=18.431, 95%CI= 2.464-137.855, p=0.00001). These indicate possible role for disease.
